Spot colours are pre-mixed colours for printing- so you can't just make up a colour like you can on CMYK.. they have a bajillion though so it is not really a problem. It really just refers to the style of printing. You know how on home printers what you print can look different from one printer to the next? That's cause it mixes the colours during the printing process with Cyan Magenta Yellow and Black (K). Professional printers use pre-mixed colours so that a document will come out exactly as the designer created it.
